Tat-braftide是一种BRAF激酶抑制剂,由10肽抑制剂braftide-TRHVNILLFM组成,由PEG连接到细胞穿透序列Tat。Tat-braftide被设计成与BRAF激酶结构域的二聚体界面结合,因此它通过破坏BRAF二聚体来抑制BRAF蛋白激酶活性,在激酶分析中IC50为43 nM。此外,以BRAF激酶的二聚体界面为靶点导致RAF和MEK的蛋白质降解,揭示了RAF在保护大型MAPK复合物免受蛋白质降解方面的新型支架功能。Tat-braftide在多种显示RAS依赖性或独立性的细胞中显示出对WT-BRAF和BRAFG469A的有效抑制和降解。Tat公司−braftide治疗导致HCT116和HCT-15细胞的生长受到剂量依赖性抑制,EC50值分别为7.1和6.6μM,表明KRAS突变的结肠癌细胞的细胞活力受到了有效抑制。
