The 162-46 monoclonal antibody recognizes human Trop-2 which is also known as GA733-1. Trop-2 is encoded by TACSTD2 (tumor-associated calcium signal transducer 2). It is a single-pass type-1 transmembrane glycoprotein. Trop-2 was originally identified to be expressed on human trophoblast and choriocarcinoma cell lines. Trop-2 is expressed in most human carcinomas. Trop-2 appears to be involved in the transduction of an intracellular calcium signal, although the exact physiological mechanism has yet to be elucidated.

Flow cytometric analysis of Trop-2.  MCF7 cells (Human breast adenocarcinoma; ATCC HTB-22) were incubated with 0.25 µg of the Purified Mouse Anti-Human Trop-2 antibody (clone 162-46; shaded) or with a mouse IgG1 isotype control (Cat. No. 555746; unshaded) in BD Pharmingen™ Stain Buffer (Cat. No.554656) for 1 hr on ice. Cells were washed twice in Stain Buffer and incubated with 1 µg of a FITC Goat Anti-Mouse Ig secondary antibody (Cat. No.554001) in Stain Buffer for 1 hr on ice. Cells were washed twice and analyzed by flow cytometry.
